I don't know, after 5 years of punching a heavy bag I suppose pain suppression is built up.
Then again, people feel pain differently, and some have damaged nerve endings.
The most painful point I know is at the base of the jaw. If you go right underneath where the jaw joins the skull (below the ear) push your thumb in and twist. On an opponent it works best with both thumbs, one on either side of the head. I don't know anyone who's had that move properly applied to them who could hold out.
Works best if your opponent is a quadriplegic

An opponent in a real fight will have heightened pain resistance due to adrenalin and obviously won't stand there.
"You" will have an enormous loss of fine motor control to effect these techniques due to adrenalin.
Nuts, eyes, throat or better yet run.
The ones on the hands do not hurt...the ones on the wrists don't hurt. However the ones near the elbows succeed in causing pain.

I've found that pressure points especially are really variable from person to person. I have friends on whom you could dig into a point for hours with no result except a bruise, and others that if you barely tap the same spot they scream in agony.
So... they're nice when they work, I guess, but definitely not anything that I'd rely on. The ones that force the associated muscle to relax seem to be more commonly effective, even if they don't necessarily hurt.
I think it goes without saying that I'm fuckin' subscribing to this mofo's channel.
